"uuid:ce0550fc-899c-47d7-87d9-9fb6f9037223","http://resolver.tudelft.nl/uuid:ce0550fc-899c-47d7-87d9-9fb6f9037223","Push tows in canals","Koster, J.","Rijkswaterstaat","1975","This publication summarizes the research carried out in the Netherlands in the 1960s into the dimensions required for the cross-section of ship canals if push tows are to navigate safely with other types of inland shipping.The necessary cross-section dimensions were determined by the space required for the different traffk situations which must be possible without danger. The traffk situation requiring the greatest amount of space - taken as the key traffk situation - involves three large vessels: one is overtaking another, while a third is sailing in the opposite direction. At first sight it would seem that trafik problems in fairways can be approached by methods similar to those used in the case of roads, i.e. by allocating a lane of a given width to each of the vessels involved. This method has been used in some previous studies. Closer consideration shows, however, that it is unsatisfactory because in a fairway not only the vessel but also the supporting environment is in motion. As a result, the transverse movements of a vessel are more difficult to control than those of a vehicle on a highway whereas in addition, the different vessels infiuence one another. As long as a single ship is sailing in the axis of a straight canal with a prismatic cross-section, it is possible to derive approximate theoretica1 values for at least the principal characteristic parameters of the water movement (drop of the water level and reverse flow). In this case sufficient data is also available from practical measurements and laboratory tests to determine with reasonable accuracy the deviations from the theoretical results as a consequence of the approximations used. However, when the ship is not navigating in the canal axis and in particular when several ships are present alongside each other in the canal simultaneously, the water movement becomes too complicated for a theoretical approximation. It is therefore still impossibIe to obtain rules for the design of a canal cross-section by theoretical means. In deze nota is enkel ingegaan op het aspekt ""benodigde hoogte"" van de zeewering. Er zijn meer kriteria waaraan een zeewering dient te voldoen, zoals: standzekerheid, piping, grondmechanische stabiliteit, stabiliteit van de bekleding, etc. 2. De gehanteerde randvoorwaarden en methoden om deze door te rekenen naar de uiteindelijke zeewering te Harlingen zijn behept met een zekere mate van onnauwkeurigheid. Met deze onnauwkeurigheid wordt rekening gehouden door onderbouwende probabilistische berekeningen te maken waarin deze onnauwkeurigheden zijn gemodelleerd. Het resultaat van de probabilistische berekeningen laat zien dat: a. met een deterministische berekening een goede verwachtingswaarde kan worden berekend van de hoeveelheid overslag. b. een indruk kan worden verkregen van de overschrijdingskans van een hoeveelheid overslag groter dan het bergend vermogen van de binnenhaven. 3. Er worden twee alternatieven gepresenteerd, welke zich van elkaar onderscheiden door de gehanteerde hoogte van de zeewering voor het onderdeel ""Keersluis"". Deze alternatieven worden gepresenteerd in tabel 1 (par. 11). Hierin staat voor elk onderdeel van de zeewering: a. De bijdrage aan de totale overslag in [m3] aan de binnenhaven. b. Het maximale gemiddelde overslagdebiet in [1/ms]. Het totaal van de belasting op de binnenhaven wordt eveneens voor elk alternatief gegeven. De hoogte van de keersluis in alternatief 1, vormt één van de zwakke schakels in het geheel. De huidige hoogte van de keersluis bedraagt: NAP +5,90 [m]. In alternatief 1 wordt een hoogte van de keersluis aangenomen van NAP + 6,40[m]. Het bergend vermogen van de binnenhaven bedraagt ca. 25.000 [m3]. Uit tabel 1 blijkt dat alternatief 1 een belasting op de binnenhaven geeft van ca. 21.000 [in3]. Dit betekent dat onder de maatgevende stormvloedomstandigheid de totale hoeveelheid overslag geringer is dan het bergend vermogen van de binnenhaven. De bijdrage aan de totale overslag door de keersluis bedraagt ca. 7.000 [m3]. De maximale hoeveelheid overslag bedraagt voor de keersluis: ca. 60 [1/ms]. Er wordt opgemerkt dat het wellicht mogelijk is een hoeveelheid water uit de binnenhaven af te laten op de boezem. Hier is geen rekening mee gehouden. 4. In alternatief 2 wordt een hoogte van de keersluis aangenomen van NAP +6,90 [m]. De berekeningen laten in dat geval zien dat de totale hoeveelheid overslag gereduceerd wordt tot ca. 15.000 [m3]. De bijdrage aan de totale overslag door de keersluis bedraagt ca. 1.000 [m3]. De maximale hoeveelheid overslag bedraagt voor de keersluis: ca. 7 [1/ms]. Een tweede zwakke schakel in het geheel wordt gevormd door het onderdeel ""doorgangen"". De hoogte van dit onderdeel is in deze nota een randvoorwaarde. Het berekende maximale gemiddelde overslagdebiet voor dit onderdeel bedraagt ca. 45 [1/ms]. Deze hoeveelheid overslag dient wel op adequate wijze te kunnen worden afgevoerd. Een alternatief, dat hier niet verder is uitgewerkt, is om dit constructieonderdeel zodanig aan te passen dat onder stormvloedomstandigheden de kerende hoogte tijdelijk wordt verhoogd. Dit kan worden gerealizeerd door middel van aan te brengen schotten in daarvoor bestemde sponningen. 5. De gepresenteerde alternatieven zijn vanuit technisch oogpunt gezien acceptabel. In geval echter de maximale hoeveelheid overslag groter is dan 10 [1/ms], is het noodzakelijk om maatregelen te treffen met betrekking tot de overslagbestendigheid van die onderdelen. By preventing religation, these drugs also prolong the lifetime of the covalent TopIB–DNA complex. Here, we use magnetic tweezers to measure the rate of supercoil removal by drugbound TopIB at a single DNA sequence in real time. This is accomplished by covalently linking camptothecins to a triple helix-forming oligonucleotide that binds at one location on the DNA molecule monitored. Surprisingly, we find that the DNA dynamics with the TopIB–drug interaction restricted to a single DNA sequence are indistinguishable from the dynamics observed when the TopIB–drug interaction takes place at multiple sites. Specifically, the DNA sequence does not affect the instantaneous supercoil removal rate or the degree to which camptothecins increase the lifetime of the covalent complex. Direct access to digitized images has revolutionized the use of electron microscopy, sparking developments such as automated collection of tomographic data, focal series, random conical tilt pairs and ultralarge single-particle data sets. Nevertheless, for ultrahigh-resolution work photographic plates are often still preferred. In the ideal case, the quality of the recorded image of a vitrified biological sample would solely be determined by the counting statistics of the limited electron dose the sample can withstand before beam-induced alterations dominate. Unfortunately, the image is degraded by the non-ideal point-spread function of the detector, as a result of a scintillator coupled by fibre optics to a CCD, and the addition of several inherent noise components. Different detector manufacturers provide different types of figures of merit when advertising the quality of their detector. It is hard for most laboratories to verify whether all of the anticipated specifications are met. In this report, a set of algorithms is presented to characterize on-axis slow-scan large-area CCD-based TEM detectors. These tools have been added to a publicly available image-processing toolbox for MATLAB. Three in-house CCD cameras were carefully characterized, yielding, among others, statistics for hot and bad pixels, the modulation transfer function, the conversion factor, the effective gain and the detective quantum efficiency. These statistics will aid data-collection strategy programs and provide prior information for quantitative imaging. The studies cover (1) current and potential benefits of bio-based products in extended value chains including activities after end-of-life of products, (2) value communication between stakeholders in extended value chains, and (3) creating an integrated development approach for optimized bio-based products. Most existing models for value chains were found to be one-way single-flow models to which iterations and interactions have been added to obtain realistic representations of best practices. Interviews of thirteen professionals clarified some barriers between the current implementation of bio-based plastics and the ideal sustainable value chain fulfilment. The most noticeable barriers included trustful information sharing, information and knowledge gaps, sub-optimum application of bio-based plastics, and the need for facilitating product design. The main designers' needs are increased integrated literacy related to bio-based materials, eco-effeetiveness and a more effective communication competence in value chains. Based on all findings, six approaches of design tasks integration into value chains have been proposed. Hydrological education that was traditionally teacher centred, where the students look up to the teacher for expertise and information, should change to better prepare hydrologists to develop new knowledge and apply it in new contexts. An important first step towards this goal is to change the concept of education in the educators' minds. The results of an investigation to find out whether didactic training influences the beliefs of hydrology educators about their teaching styles is presented. Faculty of UNESCO-IHE has been offered a didactic certification program named university teaching qualification (UTQ). The hypothesis that UTQ training will significantly alter the teaching style of faculty at UNESCO-IHE from expert/formal authority traits towards facilitator/delegator traits was tested. A first survey was conducted among the entire teaching staff (total 101, response rate 58%). The results indicated that there are significantly higher traits of facilitator and delegator teaching styles among UTQ graduates compared to faculty who were not significantly trained in didactics. Traditional thermo-mechanical recycling causes reduction of mechanical properties for most thermoplastics. Down-cycled materials may nevertheless be suited for certain useful applications. Developing such applications may be a step towards more effective future use of resources. Increased application of recycled thermoplastics is generally preferable over landfills, or waste lying around in some parts of the world. Different recycled high-impact polystyrenes (HIPS) from TVs were mechanically tested, as well as mixtures of them. Mixtures of a low-density polyethylene (LDPE) and a polypropylene (PP) were tested as well, representing a large portion of mixed plastics waste. A contribution was made to a process improvement for creating products from street waste in Kenya. Mixing different recycled HIPS grades gave no significant changes in melt flow index and impact strength. One mixture showed a significant reduction in tensile strength. In PP-LDPE mixtures there was an indication of improved mechanical impact resistance of the PP by addition of LDPE in several percentages. Therefore, new methods for improving the yield are emerging, including inspection and review methods with sufficient resolution and throughput. Existing industrial tools cannot anymore fulfill these requirements for upcoming smaller and 3D features, since they are performing at the edge of their performance. Scanning probe microscopy (SPM) has the ability to accurately measure dimensions in the micrometer to nanometer scale. Examples of applications are surface roughness, channel height and width measurement, defect inspection in wafers, masks and flat panel displays. In most of these applications, the target area is very large, and, therefore, the throughput of the measurement plays an important role in the final production cost. Single SPM has never been able to compete with other inspection systems in terms of measurement speed, thus has not fulfilled the industry needs in throughput and cost. Further increase of the speed of the single SPM helps, but it still is far from the required throughput and, therefore, insufficient for high-volume manufacturing. Over the past three years, we have developed a revolutionary concept for a multiple miniaturized SPM heads system, which can inspect and measure many sites in parallel. The very high speed of each miniaturized SPM unit allow the user to scan many areas, each with the size of tens of micrometers, in a few seconds. These earthquakes are not tectonic but induced by natural gas production from the Groningen Gas Field. Before that time there were no earthquakes in Groningen with such magnitudes. Research conducted by the Ministry of Economic Affairs (EZ) and the State Supervision of Mines (Sodm) showed that both number and magnitude have increased in recent years. The Ministry has given priority to investigate the induced earthquake risks and effects on the most critical structures, installations and infrastructure, such as industrial plants in the Delfzijl – Eemsmond region and public buildings like hospitals and schools. Here, we explore the interaction between the predatory bacterium Bdellovibrio bacteriovorus and its prey Escherichia coli in microfabricated landscapes. We ask how fragmentation influences the prey dynamics at the microscale and compare two landscape geometries: a patchy landscape and a continuous landscape. By following the dynamics of prey populations with high spatial and temporal resolution for many generations, we found that the variation in predation rates was twice as large in the patchy landscape and the dynamics was correlated over shorter length scales.We also found that while the prey population in the continuous landscapewas almost entirely driven to extinction, a significant part of the prey population in the fragmented landscape persisted over time. We observed significant surface-associated growth, especially in the fragmented landscape and we surmise that this sub-population is more resistant to predation. The genome of the haploid laboratory strain CEN.PK113-7D contains a full complement of biotin biosynthesis genes, but its growth in biotin-free synthetic medium is extremely slow (specific growth rate [μ] ≈ 0.01 h-1). Four independent evolution experiments in repeated batch cultures and accelerostats yielded strains whose growth rates (μ ≤ 0.36 h-1) in biotin-free and biotin-supplemented media were similar. Whole-genome resequencing of these evolved strains revealed up to 40-fold amplification of BIO1, which encodes pimeloyl-coenzyme A (CoA) synthetase. The additional copies of BIO1 were found on different chromosomes, and its amplification coincided with substantial chromosomal rearrangements. A key role of this gene amplification was confirmed by overexpression of BIO1 in strain CEN.PK113-7D, which enabled growth in biotin-free medium (μ= 0.15 h-1). Mutations in the membrane transporter genes TPO1 and/or PDR12 were found in several of the evolved strains. Deletion of TPO1 and PDR12 in a BIO1-overexpressing strain increased its specific growth rate to 0.25 h-1. The effects of null mutations in these genes, which have not been previously associated with biotin metabolism, were nonadditive. However, current hybridization methods are limited by their reliance on preexisting or introduced selectable phenotypes. This study presents a high-throughput phenotype-independent method for isolation of interspecies Saccharomyces hybrids based on dual dye-staining and subsequent mating of two strains, followed by enrichment of double-stained hybrid cells from a mating population by fluorescence-activated cell sorting (FACS). Pilot experiments on intra-species mating of heterothallic haploid S. cerevisiae strains showed that 80% of sorted double-stained cells were hybrids. The protocol was further optimized by mating an S. cerevisiae haploid with homothallic S. eubayanus spores with complementary selectable phenotypes. In crosses without selectable phenotype, using S. cerevisiae and S. eubayanus haploids derived from laboratory as well as industrial strains, 10 to 15% of double-stained cells isolated by FACS were hybrids. When applied to rare mating, sorting of double-stained cells consistently resulted in about 600-fold enrichment of hybrid cells. Mating of dual-stained cells and FACS-based selection allows efficient enrichment of interspecies Saccharomyces hybrids within a matter of days and without requiring selectable hybrid phenotypes, both for homothallic and heterothallic strains. Absorption of light at 700 nm leads to the almost quantitative generation of triplet excited states by intersystem crossing. In a PVE, individuals select their preferred portfolio of government projects given a constrained public budget. Individuals’ preferences for (the impacts of) government projects can be determined based on these choices. The obtained preferences can be used to rank government projects in terms of their desirability. Cost-Benefit Analysis (CBA) is an alternative appraisal method used to assess the desirability of government projects. CBA establishes the desirability of public projects through analyzing people's trade-offs between their private income and impacts of public projects. The primary objective of this paper is to investigate whether CBA and PVE lead to different policy recommendations in the context of urban mobility investments. First, this paper aligns PVE with the traditional Kaldor-Hicks welfare economics framework underlying many Cost-Benefit Analyses. Second, this paper positions PVE against other valuation methods. Third, this paper applies the PVE method to evaluate the impacts of projects mitigating flood risks in the Netherlands. Coupling of these Class-II DHODHs to mitochondrial respiration makes their in vivo activity dependent on oxygen availability. Saccharomyces cerevisiae and closely related yeast species harbor a cytosolic Class-I DHOD (Ura1) that uses fumarate as electron acceptor and thereby enables anaerobic pyrimidine synthesis. Here, we investigate DHODs from three fungi (the Neocallimastigomycete Anaeromyces robustus and the yeasts Schizosaccharomyces japonicus and Dekkera bruxellensis) that can grow anaerobically but, based on genome analysis, only harbor a Class-II DHOD. Results: Heterologous expression of putative Class-II DHOD-encoding genes from fungi capable of anaerobic, pyrimidine-prototrophic growth (Arura9, SjURA9, DbURA9) in an S. cerevisiae ura1Δ strain supported aerobic as well as anaerobic pyrimidine prototrophy. A strain expressing DbURA9 showed delayed anaerobic growth without pyrimidine supplementation. Adapted faster growing DbURA9-expressing strains showed mutations in FUM1, which encodes fumarase. GFP-tagged SjUra9 and DbUra9 were localized to S. cerevisiae mitochondria, while ArUra9, whose sequence lacked a mitochondrial targeting sequence, was localized to the yeast cytosol. Experiments with cell extracts showed that ArUra9 used free FAD and FMN as electron acceptors. Expression of SjURA9 in S. cerevisiae reproducibly led to loss of respiratory competence and mitochondrial DNA. A cysteine residue (C265 in SjUra9) in the active sites of all three anaerobically active Ura9 orthologs was shown to be essential for anaerobic activity of SjUra9 but not of ArUra9. Conclusions: Activity of fungal Class-II DHODs was long thought to be dependent on an active respiratory chain, which in most fungi requires the presence of oxygen. By heterologous expression experiments in S. cerevisiae, this study shows that phylogenetically distant fungi independently evolved Class-II dihydroorotate dehydrogenases that enable anaerobic pyrimidine biosynthesis. We studied balance robustness and performance, H-reflex gains, paired reflex depression, and co-contraction duration in ankle muscles after one and ten training sessions in 22 older adults (+65 yrs). Mediolateral balance robustness, time to balance loss in unipedal standing on a platform with decreasing rotational stiffness, improved (33%) after one session, with no further improvement after ten sessions. Balance performance, absolute mediolateral center of mass velocity, improved (18.75%) after one session in perturbed unipedal standing and (18.18%) after ten sessions in unperturbed unipedal standing. Co-contraction duration of soleus/tibialis anterior increased (16%) after ten sessions. H-reflex gain and paired reflex depression excitability did not change. H-reflex gains were lower, and soleus/tibialis anterior co-contraction duration was higher in participants with more robust balance after ten sessions, and co-contraction duration was higher in participants with better balance performance at several time-points. Changes in robustness and performance were uncorrelated with changes in co-contraction duration, H-reflex gain, or paired reflex depression. In older adults, balance robustness improved over a single session, while performance improved gradually over multiple sessions. Due to the inherent complexity and limited technical accessibility of the mitochondrial transcriptome, fundamental questions regarding mitochondrial gene expression and splicing remain unresolved, even in the model eukaryote Saccharomyces cerevisiae. Long-read sequencing could address these fundamental questions. Therefore, a method for the enrichment of mitochondrial RNA and sequencing using Nanopore technology was developed, enabling the resolution of splicing of polycistronic genes and the quantification of spliced RNA. This method successfully captured the full mitochondrial transcriptome and resolved RNA splicing patterns with single-base resolution and was applied to explore the transcriptome of S. cerevisiae grown with glucose or ethanol as the sole carbon source, revealing the impact of growth conditions on mitochondrial RNA expression and splicing. This study uncovered a remarkable difference in the turnover of Group II introns between yeast grown in either mostly fermentative or fully respiratory conditions. Whether this accumulation of introns in glucose medium has an impact on mitochondrial functions remains to be explored. Methods Overall, 87 patients undergoing total hip arthroplasty (THA) were randomized to either a Trident II HA or Trident HA shell, each cementless with clusterholes and HA-coating. All components were used in combination with a cemented Exeter V40 femoral stem. Implant migration was measured using radiostereometric analysis (RSA), with radiographs taken within two days of surgery (baseline), and at three, 12, and 24 months postoperatively. Proximal acetabular component migration was the primary outcome measure. Clinical scores and patient-reported outcome measures (PROMs) were collected at each follow-up. Results Mean proximal migrations at three, 12, and 24 months were 0.08 mm (95% confidence interval (CI) 0.03 to 0.14), 0.11 mm (95% CI 0.06 to 0.16), and 0.14 mm (95% CI 0.09 to 0.20), respectively, in the Trident II HA group, versus 0.11 mm (95% CI 0.06 to 0.16), 0.12 mm (95% CI 0.07 to 0.17), and 0.14 mm (95% CI 0.09 to 0.19) in the Trident HA group (p = 0.875). No significant differences in translations or rotations between the two designs were found in any other direction. Clinical scores and PROMs were comparable between groups, except for an initially greater postoperative improvement in Hip disability and Osteoarthritis Outcome Symptoms score in the Trident HA group (p = 0.033). Conclusion The Trident II clusterhole HA shell has comparable migration with its predecessor, the Trident hemispherical HA cluster shell, suggesting a similar risk of long-term aseptic loosening.
