Electron transport within living cells is essential for energy conservation in all respiring and photosynthetic organisms. While a few bacteria transport electrons over micrometer distances to their surroundings, filaments of cable bacteria are hypothesized to conduct electric currents over centimeter distances. We used resonance Raman microscopy to analyze cytochrome redox states in living cable bacteria. Cable-bacteria filaments were placed in microscope chambers with sulfide as electron source and oxygen as electron sink at opposite ends. Along individual filaments a gradient in cytochrome redox potential was detected, which immediately broke down upon removal of oxygen or laser cutting of the filaments. Without access to oxygen, a rapid shift toward more reduced cytochromes was observed, as electrons were no longer drained from the filament but accumulated in the cellular cytochromes. These results provide direct evidence for long-distance electron transport in living multicellular bacteria.

The advantage of using non-invasive imaging such as optical coherence tomography (OCT) to asses material properties from deformed biofilm geometries can be compromised by the assumptions made on fluid forces acting on the biofilm. This study developed a method for the determination of elastic properties of biofilms by modelling the biofilm deformation recorded by OCT imaging with poroelastic fluid-structure interaction computations. Two-dimensional biofilm geometries were extracted from OCT scans of non-deformed and deformed structures as a result of hydrodynamic loading. The biofilm geometries were implemented in a model coupling fluid dynamics with elastic solid mechanics and Darcy flow in the biofilm. The simulation results were compared with real deformed geometries and a fitting procedure allowed estimation of the Young's modulus in given flow conditions. The present method considerably improves the estimation of elastic moduli of biofilms grown in mini-fluidic rectangular channels. This superior prediction is based on the relaxation of several simplifying assumptions made in past studies: shear stress is not anymore taken constant over the biofilm surface, total stress including also pressure is accounted for, any biofilm shape can be used in the determinations, and non-linear behavior of mechanical properties can be estimated. Biofilm elastic moduli between 70 and 700 Pa were obtained and biofilm hardening at large applied stress due to increasing flow velocity was quantified. The work performed here opens the way for in-situ determination of other mechanical properties (e.g., viscoelastic properties, relaxation times, plastic yields) and provides data for modelling biofilm deformation and detachment with eventual applications in biofilm control and removal strategies.

This study focused on characterizing the structure of biofilms developed on carriers used in lab-scale moving bed biofilm reactors. Both light microscopy (2D) and optical coherence tomography (OCT) were employed to track the biofilm development on carriers of different geometry and under different aeration rates. Biofilm structure was further characterized with respect to average biofilm thickness, biofilm growth velocity, biomass volume, compartment filling degree, surface area, etc. The results showed that carriers with a smaller compartment size stimulated a quick establishment of biofilms. Low aeration rates favored fast development of biofilms. Comparison between the results derived from 2D and 3D images revealed comparable results with respect to average biofilm thickness and compartment filling degree before the carrier compartments were fully willed with biomass. However, 3D imaging with OCT was capable of visualizing and quantifying the heterogeneous structure of biofilms, which cannot be achieved using 2D imaging.@en

The bubble characteristics in a 3-D cylindrical fluidized bed have been investigated both experimentally and numerically. Experiments were performed on a 0.1 m diameter fluidized bed, with alumina oxide particles (diameter ~1 mm) as a fluidizing material. Measurements were done at a spatial resolution of 1 mm and a temporal resolution of 1000 cross-sectional images per second, using an ultrafast electron beam X-ray computed tomography (XRT) setup (Fischer and Hampel 2010). A two-fluid model using kinetic theory of granular flow (Verma et al., 2013) was used to predict the bed dynamics numerically. The equivalent bubble diameter as a function of height is in close agreement with Darton et al. (1977) and Werther (1975) correlations. The bubble size distribution predicted from simulations is broader compared to experiments. Both the bubble rise velocity and the bubble size increase with increase in excess gas velocity. The experimental measurements and simulation predictions are in fair agreement with the Hilligardt and Werther (1986) correlation.

Bubble characteristics in a three-dimension gas-fluidized bed (FB) have been measured using noninvasive ultrafast electron beam X-ray tomography. The measurements are compared with predictions by a two-fluid model (TFM) based on kinetic theory of granular flow. The effect of bed material (glass, alumina, and low linear density polyethylene (LLDPE), dp ~1 mm), inlet gas velocity, and initial particle bed height on the bubble behavior is investigated in a cylindrical column of 0.1-m diameter. The bubble rise velocity is determined by cross correlation of images from dual horizontal planes. The bubble characteristics depend highly upon the particle collisional properties. The bubble sizes obtained from experiments and simulations show good agreement. The LLDPE particles show high gas hold-up and higher bubble rise velocity than predicted on basis of literature correlations. The bed expansion is relatively high for LLDPE particles. The X-ray tomography and TFM results provide in-depth understanding of bubble behavior in FBs containing different granular material types.@en
