Abstract

This thesis describes the use of simulated moving bed (SMB) chromatography for separation of proteins. The underlying aim is the increase of the efficiency of process chromatography in terms of a reduction of the sorbent and buffer consumption in comparison to fixed bed chromatography. Both size exclusion and ion exchange separations of synthetic mixtures of proteins are considered. The main topics covered are the selection of the liquid to sorbent flow rate ratios, mass transfer effects and the implications thereof, and the development of salt gradient ion exchange SMB.
