Antimicrobial resistance (AMR) is among the top 10 public health threats, with nearly 5 million deaths in 2019 linked to AMR-related bacterial infections. (1) A One Health approach is needed to combat AMR.

Healthcare-based surveillance (HBS) of AMR provides incomplete information about the scope of the AMR threat. HBS screens only patients seeking medical attention, lacking community-level representativeness, and suffers from underreporting. (2) Consequently, researchers are turning to wastewater-based surveillance (WBS) to complement HBS. (3) WBS can provide information about AMR circulating within communities and hospitals, offering a comprehensive understanding of AMR prevalence. However, the surveillance targets and data obtained from WBS are distinct from those derived from HBS, creating uncertainty regarding their utility to the public health sector and ability to yield policy relevant information. In May 2024, participants in a workshop during the 7th Environmental Dimension of Antimicrobial Resistance (EDAR7) conference (Montréal, Canada) sought to answer four questions aimed at advancing the policy relevance of AMR data generated by WBS.

Abstract: Quantum information processing and quantum sensing is a central topic for researchers who are part of the Materials Research Society and the Quantum Staging Group is providing leadership and guidance in this context. We convened a workshop before the 2022 MRS Spring Meeting and covered four topics to explore challenges that need to be addressed to further promote and accelerate the development of materials with applications in quantum technologies. Rising seas coupled with ever increasing coastal populations present the potential for significant social and economic loss in the 21st century. Relatively short records of the full multidimensional space contributing to total water level coastal flooding events (astronomic tides, sea level anomalies, storm surges, wave run-up, etc.) result in historical observations of only a small fraction of the possible range of conditions that could produce severe flooding. The Time-varying Emulator for Short- and Long-Term analysis of coastal flood hazard potential is presented here as a methodology capable of producing new iterations of the sea-state parameters associated with the present-day Pacific Ocean climate to simulate many synthetic extreme compound events. The emulator utilizes weather typing of fundamental climate drivers (sea surface temperatures, sea level pressures, etc.) to reduce complexity and produces new daily synoptic weather chronologies with an auto-regressive logistic model accounting for conditional dependencies on the El Niño Southern Oscillation, the Madden-Julian Oscillation, seasonality, and the prior two days of weather progression. Joint probabilities of sea-state parameters unique to simulated weather patterns are used to create new time series of the hypothetical components contributing to synthetic total water levels (swells from multiple directions coupled with water levels due to wind setup, temperature anomalies, and tides). The Time-varying Emulator for Short- and Long-Term analysis of coastal flood hazard potential reveals the importance of considering the multivariate nature of extreme coastal flooding, while progressing the ability to incorporate large-scale climate variability into site specific studies assessing hazards within the context of predicted climate change in the 21st century.

Skin-targeting microscale medical devices are becoming popular for therapeutic delivery and diagnosis. We used cryo-SEM, fluorescence lifetime imaging microscopy (FLIM), autofluorescence imaging microscopy and inflammatory response to study the puncturing and recovery of human skin ex vivo and in vivo after discretised puncturing by a microneedle array (Nanopatch®). Pores induced by the microprojections were found to close by ~25% in diameter within the first 30 min, and almost completely close by ~6 h. FLIM images of ex vivo viable epidermis showed a stable fluorescence lifetime for unpatched areas of ~1000 ps up to 24 h. Only the cells in the immediate puncture zones (in direct contact with projections) showed a reduction in the observed fluorescence lifetimes to between ~518–583 ps. The ratio of free-bound NAD(P)H (α1/α2) in unaffected areas of the viable epidermis was ~2.5–3.0, whereas the ratio at puncture holes was almost double at ~4.2–4.6. An exploratory pilot in vivo study also suggested similar closure rate with histamine administration to the forearms of human volunteers after Nanopatch® treatment, although a prolonged inflammation was observed with Tissue Viability Imaging. Overall, this work shows that the pores created by the microneedle-type medical device, Nanopatch®, are transient, with the skin recovering rapidly within 1–2 days in the epidermis after application.
